Cell Proliferation Curve of Mouse ES D3 Strain Cells

The following figure compares the proliferation of 14th generation D3 mouse ES cells in our StemSure medium and in a product offered by one of our competitors (A). According to the results, both types of media exhibited similar growth performance. In addition, we found that media treated with MTG and 2ME displayed comparable cell proliferation

properties, confirming that MTG can be a viable alternative to 2ME.

Population Doubling Level of Mouse ES D3 Strain Cells

The following figure shows the relation between growth time and population doubling level when culturing several generations of D3 mouse ES cells in a StemSure medium and in a product offered by a competitor (A). The results show that an equivalent number of colonies grew in both types of media.

Colony Morphology and Expression of Undifferentiation Markers of Mouse ES D3 Cells

We cultured D3 mouse ES cells in a StemSure medium and observed colonies with a distinctive shiny appearance unique to mouse ES cells. We also found that the colonies were positive for various markers for undifferentiation.

Human iPS Cell Culture

We confirmed that the 201B7 line of human iPS cells will grow in StemSure media. Followings are the morphology and the cells to be positive for various markers indicating morphology and undifferentiation.

Cryopreservation of Mouse ES D3 Strain Cells

A serum-free cryopreservation solution for mouse ES cells and various cell lines. Mouse ES D3 strain cells were cryopreserved using StemSure Freezing Medium. The cell viability of thawed cells was measured. And then, colony formation and undifferentiated state of the thawed cells were confirmed after 10 day cultivating.

StemSure Serum Replacement (SSR)

This serum-free medium is particularly optimized to grow ES and iPS cells in culture. The composition of this product, which has not been designated as a poison, is publically available.

 

Population Doubling Level of Mouse ES D3 Strain Cells

The following figure shows the relation between growth time and colony number when culturing several generations of D3 mouse ES cells in a StemSure medium and in a product offered by a competitor (A). The results show that an equivalent number of colonies grew in both types of media.

NeuroSure Supplement Reagent for Neural Cell Culture

NeuroSure Supplement is a serum‐free supplement for neural cell culture. It is applicable to cultivate of rat hippocampal neurons and neural stem cells.


Rat hippocampal neural cells from E19 fetal rat were cultured in a basal medium with 2% NeuroSure Supplement on poly‐L‐lysine coated plates.

The rat hippocampal neurons were stained for β‐III tubulin (TuJ1, green) and for nuclei with DAPI (blue).

N2 Supplements

N2 Supplement is used as a replacement of general blood serum of neural cell culturing. It is suitable for culturing of primary nerve cell and neural stem cell. Differentiation induction of neural stem cell is triggered by element contained in FBS.

Serum substitutes like N2 Supplement are used for maintaining undifferentiated stage of stem cells during culturing.

Two kinds of N2 Supplements are available: one is prepared using transferrin (apo) and another is prepared using transferrin (holo). Transferrin (holo) is used generally but addition of N2 supplement containing transferrin (apo) may improve proliferation of neural stem cells depending on cell species.

Extracellular Matrices

It has been shown that the extracellular matrices support normal cell functions and play an important role in cell division and differentiation. These products can be used as coating agents for cell cultures.

11-1.

Laminin

A major component of the basal lamina (i.e., one of the basement membranes), laminins are relatively large trimeric proteins weighing about 900 kDa and made from three different peptide chains. These molecules are bioactive in promoting axon growth in neurons as well as general cell adhesion, migration, proliferation and differentiation.

Fibronectins

These heavy glycoproteins weighing about 250 kDa exist as polypeptide dimers and are one of the building blocks of the extracellular matrix. The RGD sequence (Arg-Gly-Asp) in the molecule allows fibronectin to bind to and interact with specific integrins on the cell surface. Fibronectins promote cell adhesion, migration and phagocytosis.

Adhesamine

This low molecular weight molecule has been found to promote cell proliferation and adhesion.

Adhesamine has been observed to facilitate the attachment of suspended cells to the containers in which they are being cultured in an action similar that of fibronectin. The cells can be detached by replacing the medium and washing away the adhesamine. This molecule is known to be effective for HeLa, HEL293, CHO and mouse ES cells1).

Collagens

Proteins distributed widely throughout the bodies of vertebrates, collagen is best known as a major component of connective tissue. Not only is collagen ideal for growing first generation endothelial, skin, nerve and hepatic cells, it can also be used as a matrix for 3D cultures.

 

(1) Cellcampus (Type I-collagen extracted from fish scales)

Cellcampus is a product of Taki Chemical Co., Ltd. and collagen extracted from scales of farmed tilapia (tropicalfish). Tilapia carries no viruses common to man and collagen extracted from fish has a high denaturation temperature. These characteristics make it useful for culture in the field of regenerative medicine. Cellcampus also has high fibrotic and cell proliferative potentials that make it suitable for common cell culture.

Extracted from fish (Tilapia).

Fish-derived collagen is safe as fish carries no viruses known to infect man.

High denaturation temperature.

As tilapia is a tropical fish, the denaturation temperature of collagen extracted from it is high.

This makes the fish-derived type I collagen resistant to denaturation at normal cell culture temperature (can be used in normal collagen coated culture).
