  These kits are designed for quantitative determination of βAmyloid (1-42, x-42, 1-40, and x-40), which is correlated with Alzheimer's disease. The Human β Amyloid (1-42) ELISA Kit Wako High-Sensitive (Kit ) and the Human/Rat β Amyloid (42) ELISA Kit Wako High-Sensitive (Kit ) A (40) and A (x-40) use Fab' fragments of the monoclonal antibody BC05, which recognizes the C-terminal portion, while the Human β Amyloid (1-40) ELISA Kit Wako Ⅱ (Kit ) and the Human/Rat β Amyloid (40) ELISA Kit Wako Ⅱ (Kit ) use F(ab')2 fragments of the labeled antibody BA27, which recognizes the C-terminal of Ab40 to achieve more stable antigen-antibody reactions while reducing nonspecific binding.

[Standard Curve]
  A standard curve is generated using the standard solution contained in the kit. For kits and , the standard curve was fitted as the linear relationship in the concentration range of 0.1 to 20.0 pmol/L.
For kits and , the standard curve was fitted as the linear relationship in the concentration range of 1 to 100 pmol/L.

[Application Data] Human and Mouse Plasma
  Blood was collected using vacuum blood collection tubes containing EDTA-2K. Plasma was separated by centrifugation at 5000 x g for 15 minutes at 4öC and stored at -80öC until used. The plasma sample was diluted four-fold with Standard Diluent in the kit and measured.

(Aβ(x-40), Aβ(1-40), Aβ(x-42), Aβ(1-40), and Aβ(1-42) were measured by kits 4, 3, 2, and 1, respectively

β-Secretase Inhibitors suitable to in vivo research

  Amyloid beta protein (Aβ) is a possible cause of Alzheimer's disease. The Aβ protein is generated from amyloid precursor protein (APP) by β-secretase (β-site APP Cleaving Enzyme; BACE) and gamma-secretase-mediated protease cleavage. The Aβ protein typically exists as either a 40 or 42 amino acid residue.
This product is a β-secretase inhibitor developed Yoshiaki Kiso, PhD, Nagahama Institute of Bio-Science and Technology. This inhibitor has low molecular weight and enhanced cell membrane permeability and blood-brain barrier permeability. Molecular weight reduction was achieved based on the amino acid sequence of the Swedish mutant amyloid precursor protein (APPsw) commonly observed in familial Alzheimer's disease. Both peptide and non-peptide products are available. Please use this product for research in the pathogenetic mechanism of Alzheimer's disease.


  KMI-429 is a BACE1 inhibitor with enhanced cell membrane permeability from a reduction in the molecular weight compared with existing peptide inhibitors.

  Glial cell line-derived neurotrophic factor family is a subfamily of the TGF-beta superfamily. GDNF family is structurally characterized by seven conserved cysteine residues at the C-terminal.
GDNF, neurturin, and artemin promote the survival of dopaminergic neurons, motor neurons, sensory nerve neurons, and sympathetic neurons in various neuron culture systems. These three GDNF families work on neurons derived from peripheral ganglia. In contrast, persephin promotes survival of dopaminergic neurons and motor neurons from the central nerve system.

  Wakofs N2 Supplement is used as replacement of general blood serum of neural cell culturing. Product is suitable for culturing of primary nerve cell and neural stem cell. Differentiation induction of neural stem cells is triggered by elements in FBS. Serum substitutes like N2 Supplement are used for maintaining undifferentiated stage of stem cells during culturing.
Wakofs N2 Supplements are prepared by either using transferrin (Apo) or transferrin (Holo). We have confirmed that cell proliferation is more enhanced with addition of N2 Supplement for a certain cell types.

1. Suspend 1 to 2 x 106 cells in 1 mL of this product, place in a preservation tube.
2. Preserve at -80ºC for 1 day, then preserve at -150ºC for 2 months.

<Thawing>
3. Thaw in a hot bath at 37ºC.
4. Suspend in a culture medium and then centrifuge.
5. Apply suction to the supernatant and then suspend in culture medium.

  Mouse LIF (leukemia inhibitory factor) was found as a factor inducing differentiation of M1 cells, mouse leukemia-derived cells, to macrophages and, thereafter, its inhibitory action on differentiation of embryonic stem cells (ES cells) was reported. At present, it is used in the culture of mouse ES cells as a factor maintaining undifferentiating property of mouse ES cells.
This product is a recombinant protein prepared by expression of mouse LIF in E. coli. It contains 6 Histidine tags at the C-terminal.
StemSure series is a group of products guaranteed for cell proliferation and undifferentiation property by the practical tests (cell proliferation test or colony forming test) and alkaline phosphatase (ALP) staining using mouse ES cell D3 strain.

  This product can be used for preparation of animal models of schizophrenia. When administered to rats or other animals, this product suppresses neuropoiesis. In patients with schizophrenia, a function that filters out unnecessary information, such as neighboring noise, is impaired. This sensory filtering function can be assessed by a physiological test: Prepulse Inhibition (PPI). PPI is a phenomenon in which acoustic prestimulus attenuate the reaction to a subsequent startling acoustic stimulus. It has been reported that PPI deficits may be associated with a reduction in neuropoiesis.
