Water- and organic solvent-based eluents are frequently used for reverse phase chromatography. Particularly, as organic solvents, methanol and acetonitrile are widely used. Methanol shows high solubility to ionic samples, such as acid and basic compounds.
However, since methanol significantly changes the column pressure (viscosity) depending on the ratio of mixing with water, it is necessary to carefully check the pressure particularly in the case of gradient elution. Acetonitrile shows high solubility to polar compounds and allows elution at low column pressure. When any of these eluents is used, Wakosil® C18 Series can be used safely because they have been designed to have low pressure loss and withstand high pressure.
