PARP Inhibitors

PARP (Poly ADP-ribose Polymerase) is activated when it becomes attached to regions of damaged DNA, and is involved in posttranslational modification where it adds ADP-ribose residues to nuclear proteins. PARP-1 and PARP-2 have been reported to participate in DNA repair and regulation of cell death and differentiation.

A PARP inhibitor is thought to induce cell death in cells with mutation in breast cancer susceptibility genes (BRCA) because DNA damage caused by the inhibitor cannot be efficiently repaired.
PARP inhibitors are promising cancer drug candidates.
