DISPASE manufactured by Godo Shusei Co., Ltd. (Japan) is a metallo protease that cleaves the N-terminal peptide bonds of neutral, non-polar amino acids. It is able to ablate epithelial cells from tissues as a sheet and has been used for cell separation and dispersion for primary culture.
DISPASErarely causes cell damage and is featured by gentle cell dispersion unlike the activity of protease such as trypsin or collagenase, leading to a practical use in a new field of ES or iPS cells for cell dispersion or passage.
Recently it has been redesigned as a product without using animal derived ingredients

2. Features


  1. DISPASE is neutral metalloprotease derived from Paenibacillus sp. (original name: Bacillus polymyxa). It contains Zn2+ in the active center and the activity is stabilized by Ca2+.
  2. Enzyme activity is hardly inhibited by the components of serum and it can be used regardless of existence of serum. Enzyme reaction can be stopped by addition of EDTA or dilution of reaction solution. It has a feature to break down type IV collagen or fibronectin, which compose the basal membrane, well so that it is able to ablate epithelial cells from tissues as a sheet.
  3. It has a different action from proteases such as trypsin and collagenase, it shows less cell dysfunction and more moderate cell dispersion.
  4. Since the action condition is relatively wide-ranging, the enzyme is able to be used under various conditions such as enzyme concentration, processing time, temperature, and pH.
  5. It is stable at a general temperature for cell culture 37°C, and suspension culture is also available by adding this product depending on cell strains.
  6. Since animal derived ingredients are not used, there is no contamination of Mycoplasma.
