PURESYSTEM classic II series is a proprietary protein synthesis and purification technology which is a in vitro protein synthesizing system
"reconstituted" from translation factors expressed in Escherichia coli. It is a novel reconstituted system consisting of around 30 purified enzymes
necessary for transcription, translation and energy recycling1). The translation factors, derived from E. coli are three initiation factors (IF1, IF2 and IF3),
three elongation factors (EF-G, EF-Tu and EF-Ts), three release factors (RF1, RF2 and RF3), RRF for termination factors, 20 aminoacyl-tRNA
synthetases (ARSs), methionyl-tRNA transformylase (MTF), T7 RNA polymerase and 70S ribosomes. In addition, PURESYSTEM classic II system
contains tRNAs, NTPs, amino acids and energy recycling factors.
All the components involved in transcription and translation except for ribosomes are tagged with hexahistidine at the N or C terminus. Because all the
factors are tagged and known components, synthesized protein can be easily purified by simply removing both the ribosome by ultrafiltration and the
histidine-tagged factors and enzymes by metal affinity resin. Pure protein of interest can be synthesized and purified about 3 hours.
PURESYSTEM S-S series are kits, which components are adjusted to constitute disulfide linkage to synthesized protein.
